Laboratory Practicum for Molecular and Genetic Epidemiology

OBJECTIVES

This course introduces practical aspects of the generation of molecular and genetic data from human clinical specimens, including from blood, the oral cavity, and pathological specimens. Working in the Laboratory for Molecular Epidemiology, students will develop familiarity with procedures to obtain/process clinical specimens, prepare and isolate DNA and RNA, design and execute PCR assays, generate genotype data, execute and analyze microarrays, and write a laboratory-based protocol.

By the completion of the course, students will:

1. Demonstrate familiarity with the field and laboratory procedures for collecting, processing, and storing biological specimens such as cells, plasma, and saliva, including database tracking and quality control procedures.
2. Become familiar with genetic databases and online tools that are useful in planning and performing laboratory analyses.
3. Explain procedural methods for setting up and performing PCR reactions, including the planning (calculations), additions of DNA and reagents, inclusion of proper controls, and operation of thermocyclers.
4. Demonstrate familiarity with post-PCR analytical techniques for diagnosis of polymorphisms, including electrophoresis, sequencing, RFLP, and "taqman" procedures.
5. Develop a genotyping protocol for single nucleotide polymorphism(s) of interest and write a research protocol based on the analysis of these polymorphism(s) in their own research field.
6. Become familiar with the types and uses of microarrays and associated analytical procedures.
7. Be able to define a use for protein and antibody-based markers in your research, and the newer analytical methods (luminex, mass-spec).

PREREQUISITES

Designing Clinical Research (Epi 202) and Molecular and Genetic Epidemiology I (Epi 217). Exceptions to these prerequisites may be made with the consent of the Course Director, space permitting.
